Abstract: The genetic diversity and structure of six populations by mitochondrial COⅡ sequences. The results showed that the of topmouth culter Culter alburnus were investigated nucleotide polymorphism index of the topmouth culter ranged from 0 to 0.00444. There were six haplotypes in the six populations with diversity index of 0.6048. The population pairwise genetic divergence values changed from 0.00085 to 0. 00454. The molecular variance within and among the populations of the topmouth culter was 57.42%, and 42.58%. Thirty - six samples were divided into two major distinct clusters, one including Xingkaihu, Nanwan and Taihu populations, and the other including Sanxikou, Fuqiaohe and Liangzihu populations. This study revealed that COⅡ gene of mtDNA was unsuitable to be used as molecular marker for related studies for the analyses of population genetics, and only identified topmouth culter from relative large areas.
