Abstract: The genetic diversity and population genetic structure were investigated in 57 individuals in five geographic populations of rock worm Marphysa sanguinea collected from Dalian, Xingcheng, Rushan and Beihai by a 440 bp segment of the mitochondrial DNA COⅠ gene.In total, there were 78 polymorphic sites with 19 single variable sites,59 parsimony informative sites and nine haplotypes.The overall haplotype diversity and nucleotide diversity were 0.830 and 0.097 73 in the five populations, respectively, with the maximal genetic diversity in Rushan population and the minimal one in Dalian population.A neighbor-joining phylogenetic tree and a median-joining network revealed that most haplotypes were highly interconnected and all haplotypes clustered into two branches,displaying clear different geographical distributions in the studied area.The findings will provide useful information on biodiversity protection and artificial breeding of this species.
