饲料中不同铜源和水平对凡纳滨对虾生长、免疫和组织铜含量的影响

摘要 在凡纳滨对虾Litopenaeus vannamei的配合饲料中分别添加两种形式的铜——硫酸铜和蛋氨酸铜,添加量分别为10、30、50mg/kg,饲喂凡纳滨对虾8周,并于试验的第4周末、第8周末取对虾称重,同时采集对虾的血清、肝胰脏和肌肉,测定其血清酚氧化酶(PO)和超氧化物歧化酶(SOD)活性及肝胰脏和肌肉中铜的含量。结果表明:1)4周时,硫酸铜各组虾的增重率和对照组元显著差异,蛋氨酸铜各组虾的增重率显著高于对照组,添加量为10mg/kg时,铜源之间增重率差异极显著;8周时,硫酸铜各组虾的增重率显著高于对照组,蛋氨酸铜各组虾的增重率极显著高于对照组,添加量为30mg/kg时,铜源间增重率差异显著。2)两种铜源的各添加水平对对虾的成活率元影响。3)硫酸铜各组虾的PO、SOD值显著高于对照组,蛋氨酸铜各组虾的PO、SOD值极显著高于对照组;添加水平为10mg/kg时,两种铜源对PO和SOD的影响差异极显著;添加水平为30mg/kg时,两种铜源对SOD的影响差异显著。4)两种铜源的各添加水平对肌肉中的铜含量无显著影响;蛋氨酸铜各组虾的肝胰脏中铜含量极显著高于对照组,硫酸铜各组表现为随添加水平的增加,虾肝胰脏中铜含量极显著增加,10、30、50mg/kg添加水平下,铜源对肝胰脏铜含量有极显著影响。对对虾生长、酶活、组织中铜含量的统计结果表明:铜源之间存在显著差异,蛋氨酸铜的营养效果好于硫酸铜;饲料中蛋氨酸铜的添加量为10mg/kg时,即可满足对虾生长、免疫的需要。

Abstract: A feeding trial was conducted to investigate growth, immunity and Cu concentration in some organs of white leg shrimp Litopenaeus vannamei with initial body weight of 0.2 g fed the diets containing 10, 30, 50 mg/kg Cu either as CuSO4 (Cu) or Cu - methionine (MCu). The shrimps were randomly stocked into tanks of each 0.3 m^3 at a rate of 30 individuals per tank, fed 4 times a day at a feeding rate of 8.0% - 10% for 8 weeks and weighted at week 4 and week 8. The serum, hepatopancreas and muscle were sampled and serum phenoloxidase (PO), superoxide dismutase (SOD) and Cu content in hepatopancreas and muscles determined. The results showed that there was no significant difference in weight gain rate (WGR) at the end of 4 weeks in Cu groups. In the MCu groups, however, significantly higher WGR (P 〈0.05) was observed, especially in the shrimp fed the diets containing 10 mg/kg MCu ( MCu - 10 group). The very significantly difference in WGR was found in the two Cu sources at supplemental level of 10 mg/kg diet(P 〈 0.01 ). There was no significant difference in WGR among MCu - 10, MCu - 30 and MCu - 50. But significant differences were found between two Cu sources ( P 〈 0.05) at 30 mg/kg diet at the end of 8 weeks. Supplemental levels and Cu sources did not affected significantly the survival rates at the 4 th and 8^th weeks ( P 〉 0.05). There were significantly higher PO and SOD activities in MCu - 10, MCu-30 and MCu-50 groups than those in the Cu groups (P 〈0.01), no significant difference among MCu - 10, MCu- 30 and MCu -50 groups. The significantly differences in SOD were found between two Cu sources at supplemental level of 30 mg/kg diet ( P 〈 0.05). Cu contents in muscles were not affected by supplemental levels and Cu sources (P 〉 0.05). Cu concentration in hepatopancreas, however, was much significantly higher in MCu - 10, MCu - 30 and MCu - 50 groups than those in the Cu groups.
