Abstract: In this study,partialmitochondrial DNA Cytb gene was extracted and analyzed by a biologicalmethod for software access to design the genetic diversity and structure in four populations(Kaidu River,Manas River,Kashi River,and Gongnaisi River)of Gymnodipty chusdybowskii.The results showed that 15 variable nucleotide sites and 9 haplotypeswere detected in 74 individuals,in which 1 variable siteswas found in twenty-one individuals in Kaidu River.Ten variable sites were found in the twenty-seven individuals in Gongnaisi River,5 variable sites in the two individuals in Kashi River and 9 variable sites in the twenty-four individuals in Manasi River.G. chusdybowskii had average haplotype diversity Hdof0.810 and nucleotide diversity Piof0.005 9.The pairwise fixation index(FST)revealed that there was a high genetic differentiation between the Manas River and other groups(FST>0.15),and that a lower differentiation in the other groups(0.05<FST<0.15).Gene flow(Nm)showed that there was a great river of gene flow between populations in Kashi River and Gongnaisi River.Neighbor-joining tree and Median-joining network of G.dybowskii also indicated that Kashi River populations and other population had farther relation in haplotypes,and that therewas close significant differentiation in the other haplotypes of the other three groups.The findings showed that four groups of G.chusdybowskii germplasm resources have suffered from great damage,and that fisheriesmanagementmeasures should be strengthened to set up a special protection agency.
