Abstract: The DNAs of swimming crab Portunus trituberculatus in four geographical populations in Dalian,Weifang,Rizhao and Zhoushan were extracted,and whole length of ND5 gene(accession number:NC-005037) of mitochondrial DNA was amplified by PCR with a pair of primers designed with primer premier 5.0.The PCR products were sequenced by agr-gel and purification with TIANquick Midi Purification kit after they were tested.Then,the sequences were analyzed by Clustal X 1.83,DNAMAN,DnaSP Version 5 and MEGA 3.1 software.The PCR products covering the whole sequence of ND5 gene(1 728 bp) showed 1908 bp in length.The ND5 gene fragments had mean of 39.5% of A,31.5% of T,10.1% of G and 18.9 of % C,and 71.1% of A+T,higer than that of G+C significantly.There were 16 variation sites in those gene fragments,including one transvertion site and fifteen transition sites.There were eight haplotypes among the ten sequencing samples,and the nucleotide diversity(Pi) was 0.00289,the haplotype diversity Hd 0.924.Analysis of genetic diversity of ND5 gene in the crab provides basic information for management,protection and utilization of the economically important marine species.
