Abstract: In the present study, a pair of primer was designed and synthesized according to the full - length cDNA sequence of interleukin - 8 ( IL - 8 ) found in common carp ( Cyprinus carpio). A specific gene fragment of the IL - 8 was extracted, purified and amplified from total genomic DNA extracted from the spleen of common carp using PCR. The purified products were then inserted into pMD18 -T vector which was transformed to competent Escherichia coli DH 5or for sequencing. Results indicated that the common carp IL - 8 gene encompassed 943 nucleotides, 4 exons, and 3 introns, which was very similar to the known IL- 8 genes of other species with the same exon/intron arrangement. The splice sites had been well conserved through evolution, and observed the regulation of GT - AG completely. A phylogenetic analysis using Interleukin - 8 protein sequences from the GenBank verified the presumed orthologous relationships of the common carp gene to their mammalian counterparts, and revealed a closer relationship between common carp IL- 8 and zebrafish IL- 8 than between carp IL- 8 and mammalian IL- 8. The structure of carp IL- 8 gene has been also well conserved through rainbow trout. The base number of all exons was almost stable, evolution compared with human, pig, dog, and ahhongh the introns were more variable than exons.
