Abstract: The enzymatic hydrolysis of protein from processed scallop disposal was studied using two groups of proteases through the orthogonal tests. The optimum parameters for the mixture of two groups of proteases were: the enzymatic ratio of neutral proteinase to flavourzyme 2: 1, a temperature of 65℃, pH 7.0 and 5 hours for the mixed proteases of neutral protease and flavourzyme. The hydrolysis results under the optimum conditions showed that content of amino nitrogen in the hydrolytes was 50.8% and the degree of protein hydrolyzing in the hydrolytes was 95.8%. The optimum parameters for another mixed proteases were: the enzymatic ratio of papain to flavourzyme 2: 1, a temperature of 65℃, pH 7.0, and 5 hours. The content of amino nitrogen and the degree of protein hydrolyzed under the optimum conditions were 43.7% and 85.7%, respectively. The molecular distributions of the peptides in the hydrolytes analyzed by Sephadex G - 15 resin indicated that the peptide molecular weights hydrolyzed by two groups of mixed proteases were about 585 Da and 589 Da, respectively. These findings illustrated that the peptides in hydrolytes were mostly small molecular peptides,and can be further processed as new products of hydrolyzed protein.
