Abstract: Our previous study showed that there was no cross protection in vaccines prepared from Streptococcus agalactiaestrains with different serotypes (Ia and Ib) isolated from Nile tilapia Oreochromis niloticus. Soluble protein of Ia and Ib serotypes S.agalactiae strains were extracted, isolated and analyzed using two dimension-Difference Gel Electrophoresis (2D-DIGE) and the differential expression protein spots and the differential expression proteins were identified by MALDI-TOF-MS in order to understanding of the reason for the immunogenicity difference between Ia and Ib serotypes tilapia S.agalactiae. The functional category of the identified protein was determined by DAVID bioinformatics database. The results showed that there were 167 differential expression protein spots between these two serotypes strains in the 2D-DIGE gel and 32 proteins were identified by MALDI-TOF-MS. Most of the identified proteins were involved in related biological processes including oxidation-reduction, generation of precursor metabolites and energy and glucose metabolic process, especially involved in glycolysis/gluconeogenesis, pyruvate metabolism and fatty acid biosynthesis pathways. Protein function prediction indicated that GAPGH, cysteine synthase and manganese-dependent superoxide dismutase were associated with immunity and vaccine developing. The findings firstly approve that there are lots of differential expression proteins between Ia and Ib serotypes of S.agalactiae strains, some of which are immune-related proteins and (or) vaccine-related proteins, which is contributed to the immunogenicity different between Ia and Ib serotypes of S.agalactiae strains, but still need further research.
