SNPs screening of 3'UTR in Apoprotein A-I-1 gene and its association with growth traits in grass carp

Abstract: Screening and genotype SNPs in 3'UTR of Apoprotein A-I-1 gene were conducted in grass carp(Ctenopharyngodon idellus)through PCR sequencing and PCR-RFLP method from EST database.Two SNPs(C792T and G851A)were obtained in the cultured population of grass carp from Pearl River.The CC genotype in C792T site was found 46.53%,CT genotype 50.69%,and TT genotype 2.78%.The GG genotype in G851A was found 77.08%,GA genotype 20.83%,and AA genotype 2.08%.A general linear model was used to analyze associations of apoA-I-1 gene with 4 major growth traits(body weight,body length,body height and head length).The results showed that the mean body weight of BB genotype was higher than that of CC and CT genotype in C792T site (P>0.05).The mean weight,body length,body height and head length of GG genotype in G851A was higher than GA and AA genotype(P<0.05).Six diplotypes were constructed based on two SNPs in the experimental population.The mean body weight of diplotype D3(TTC792T-GGG851A)was higher than other diplotypes.The diplotype D6 (CCC792T-AAG851A)had the minimal mean body weight,and had significant differences in body length,body height and head length(P<0.05).TT genotype in C792T site was advantage genotype.GG genotype in G851A site was advantage genotype,and AA genotype disadvantage genotype.It is suggested that apoA-I-1 gene be a candidate modifier gene for grass carp growth traits,indicating that the apoA-I-1 gene could be useful for grass carp molecular breeding.
