Purification and structure of fucoidan from Sargasso weed Sargassum

摘要 为研究马尾藻Sargassum褐藻多糖硫酸酯的组成及结构,以马尾藻褐藻多糖硫酸酯粗品为原料,利用DEAE-Sepharose Fast Flow弱阴离子交换层析对粗品进行分离纯化,采用高效液相色谱法(HPLC)对粗品(F)及纯化后的两个组分(F0、F1)进行了单糖组成分析,同时利用红外光谱法、核磁共振波谱法研究了F0及F1的多糖结构。结果表明:用高效液相色谱法对单糖衍生物的分离效果良好,F、F0、F1均含有甘露糖、鼠李糖、葡萄糖醛酸、葡萄糖、半乳糖、木糖、岩藻糖;红外光谱分析显示,F和F0的硫酸基主要连接于岩藻糖的C2和C3位置。本研究结果可为马尾藻褐藻多糖硫酸酯的生物活性研究奠定基础。
Abstract: The fucoidan in Sargasso weed Sargassum was purified by DEAE-Sepharose Fast Flow anion-exchange chromatography and the structure characteristics of the fucoidan fractions(F0 and F1)in the crude fucoidan(F)were analyzed by infrared spectrum(IR),high performance liquid chromatography(HPLC)and nuclearmagnetic resonance(NMR)spectroscopy to evaluate themonosarccharide composition and structure of fucoidan from Sargasso weed.The HPLC revealed thatmonosaccharide composition of these fractionswere comprised ofmannose,glucuronic acid,galactose,and fucose,with a little amountof rhamnose,glucose and xylose.The IR revealed that the sulfate site of F and F0 were located at Fuc C2 and C3.The findings provide basic data for the biological activity studies of the fucoidan from Sargasso weed.
