Purification and characterization of fucoidan from brown sea weed Scytosiphon lomentaria

摘要 以从大连沿海新鲜萱藻Scytosiphon lomentaria中提取的褐藻多糖硫酸酯为原料,采用DEAE-Sepharose Fast Flow弱阴离子交换层析的方法对萱藻褐藻多糖硫酸酯粗提取物进行分离纯化,对纯化后各组分的硫酸根、总糖含量进行了测定,并进行组分分析。结果表明:各组分的硫酸根含量分别为10.64%、10.47%、11.93%、21.13%,多糖含量分别为46.56%、55.88%、50.06%、73.65%;通过单糖分析、红外光谱法和核磁共振波谱法分析,显示F-1、F-2含有鼠李糖,F-0、F-3出现岩藻糖的C6信号峰。本研究结果为下一步开发萱藻资源、了解其功效关系提供了理论基础。
Abstract: The crude fucoidan from fresh brown sea weed Scytosiphon lomentaria collected in Dalian coast was further purified by DEAE Sepharose fast flow weak anion exchange chromatography, and then four fractions, designated as F-0, F-1, F-2, and F-3, were obtained.Their sulfate contents were determined to be 10.64%,10.47%,11.93%,21.13%,and the total polysaccharide contents were analyzed to be 46.56%,55.88%,50.06%,and 73.65%,respectively.The infrared spectroscopy and nuclear magnetic resonance analysis illustrated that F-1, and F-2 contained rhamnose, and signal peaks of fucose C6 were found in F-0 and F-3.
