Expression comparisons of heat shock proteins gene in gills of zebrafish exposed to both normoxia and hypoxia

Abstract: The transcriptome analysis of heat shock proteins (HSPs) was conducted and differential expressed genes were screened in gills of zebrafishDaniorerio exposed to hypoxia and normoxia for two weeks in order to research the biological functions of zebrafish responding to the hypoxia stresses. Among the compared gill transcriptome data, 80 heat shock protein family members were acquired, and 28 differential expressed HSP genes were screened in the hypoxic and normoxic gills. It was found that 16 and 12 genes showed significantly up-regulated and down-regulated expressions(P<0.05), respectively,Hsp47 being the most up-regulated HSP family members under the hypoxia condition, andHsp90ab1 being the only up-regulated macromolecule HSP member. Gene expressions ofHsp47 andHsp90ab1 were detected in different tissues under hypoxia and normoxia by qRT-PCR analysis, indicating thatHsp47 had significantly up-regulated expressions in the gill and muscle tissues(P<0.01), and thatHsp90ab1 showed significantly up-regulated expressions in the gill, skin and muscle tissues under hypoxia stress(P<0.05 orP<0.01). The western blotting analysis revealed that HSP47 and HSP90ab1 proteins showed significantly up-regulated expressions in gills of zebrafish exposed to different hypoxia stresses(P<0.01). It is concluded that HSP47 and HSP90ab1 play important roles in hypoxia response and are the most important HSP family members during the hypoxia response.
