Effects of dietary emodin on antioxidant ability, growth and hepatopancreatic CYP450 enzyme activity in common carp Cyprinus carpio Jian

Abstract: Juvenile common carp Cyprinuscarpio Jian with initial body weight of about 150 g were reared in a circulating water system of 240 L tanks and fed basic diet (control group), and basic diets containing 30% CCl4 (model control group), and emodin (EMD)at a dose of 1 (low dose group), 3(mid- dose group) and 5 (high dose group) g/kg at water temperature of (27±2)℃ with 2 parallel fish in each group and 10 parallel fish in each group for 60 days. Then the antioxidant activity of indices, hepato-somatic index, growth performances, biochemical indices, total activity of CYP450 in hepatopancreas were determined, and CYP1A and CYP3A mRNA expression level were detected in the common carp injected with 30% CCl4to induce liver injury intraperitoneally in model control group, and in emodin supplementation groups in order to fully evaluate the liver protection effects of emodin (EMD) on aquatic animals. The results showed that EMD had a strong scavenging ability of DPPH free radicals, with scavenging ability of 81% at 6 mg/mL dietary EMD.There were significantly increase in relative weight gain rate (RWR) and specific growth rate (SGR), and decrease in food conversion ratio (FCR)in the common carp in mid- dose group and high dose group. EMD led to stabilize the liver cell membrane, to inhibit the leakages of glutamate transaminase (GOT), glutamate transaminase (GPT), and lactate dehydrogenase (LDH), and to recovery levels of Alb and total protein (TP) in serum(P<0.05). The mitigation of oxidative stress, promotion of recovery of total antioxidant capacity (T-AOC), and activities of superoxide dismutase (SOD), glutathione (GSH) and glutathione peroxidase (GSH-Px) in liver homogenate, and significant inhibition of the synthesis of malondialdehyde (MDA) were observed in the common carp fed the diets containing EMD(P<0.05). There were significant decrease in the content of CYP450 enzyme in liver and inhibition of the up-regulation of mRNA expression of CYP1A and CYP3A induced by CCl4 in common carp fed the diets containing EMD. The findings indicated that diets containing 3-5 g/kg EMD showed significant hepatoprotective effects via the anti-oxidation, scavenging free radicals and inhibiting the induction of CYP450 by CCl4.
