Effects of chitosan nanoparticles on the growth, immune enzyme, and genes of tiger puffer Takifugu rubripes

Abstract: Juvenile tiger puffer Takifugurubripes with body weight of(9.910±0.306)g were fed diets containing 0%(control group), and 0.2% of chitosan oligosaccharide(COS), 0.2% of the chitosan(CS),and 0.2%, 0.1%, and 0.02% of the chitosan nanoparticles(CN)at water temperature of 17-23 ℃ for 50 d, and activities of alkaline phosphatase(AKP), superoxide dismutase(SOD), catalase(CAT), acid phosphatase(ACP)and lysozyme(LZM)were determined and the relative expression levels of c3, hsp70, ifr2 and tlr3 genes were analyzed in liver, spleen and kidney by real-time quantitative PCR at the end of the experiment in order to investigate the effect of chitosan nanoparticles, a new immunopotentiator, on the growth performance and immune of tiger puffer.The results showed that chitosan nanoparticles led to significantly promote the growth and to increase in activities of AKP and CAT in liver, ACP and LZM in spleen, and SOD and LZM in kidney of tiger puffer(P<0.05).There were significant increase in the relative expression levels of c3, tlr3 and hsp70 genes in spleen in the tiger puffer fed the diets containing chitosan nanoparticles(P<0.05), without significant improvement of the relative expression of ifr2 gene(P>0.05).It was concluded that dietary chitosan nanoparticles resulted in improvement of the growth performance and immunity of tiger puffer, and its efficacy was equivalent with the chitosan oligosaccharide additive.
