In order to acquire hemoglobinless mutant and investigate the function of hae3 gene during hemoglobin generation,a model of hae3 gent was constructed using Cas9-CRISPR technique in zebrafish Danio rerio model which lacks of hae3 gene.Hae3 sgRNA and Cas9 RNA mixture were injected into the animal pole of zebrafish embryo with the first cleavage stage through microinjection, and identified by sequencing, multiple sequence alignment and T7 RNase digestion methods. The hemoglobin identification was conducted by O-dianisidine staining for both the hae3 knockout group and negative control. The results showed that the expression of hemoglobin was drastically decreased in the zebrafish which lacks hae3 gene and showed deformity phenotype compared with the wild type. Thus,we validate that lacking of hae3 gene severely affects the generation of hemoglobin and leads to developmental malformation during the embryonic period.

MiRNAs influencing complement C3 in sea cucumber Apostichopus japonicus were screened in the sea cucumber challenged with LPS,referenced to genome of sea urching Strongylocentrotus purpuratus,41 miRNAs by software,0,6,and 12 h after challenge by high-throughput sequencing and bioinformatics methods, and content change in related miRNAs and complement AjC3 mRNA was detected by real-time quantitative PCR to evaluate the immunity in the sea cucumber.Three miRNA(spu-miR-133,spu-miR-137 and spu-miR-2006) was shown to affect their target genes, the sea cucumber complement C3.The RT-PCR showed that the expression of spu-miR-133 was increased from 0 h to 12 h,with the peak value at 12 h,while the expression of spu-miR-137 was decreased from 0 h to 9 h and increased from 9 h to 12 h.The spu-miR-2006 had the peak expression value at 9 h and then decreased in expression. The expression of AjC3 mRNA was found to be increased from 0 h to 9 h and decreased from 9 h to 12 h.It was speculated that spu-miR-133 and spu-miR-137 inhibited the translation of complement AjC3 mRNA directly and that spu-miR-2006 regulated the expression of other genes in immune response. The findings provide a theoretical basis for the immune research.

To understand the adaptation mechanism of Schizothorax prenanti to low temperature,we designed primers based on the RNA sequences of S.prenanti and amplified the full-length coding sequence of the Schizothorax prenanti zona pellucida 3(sp-zp3) by RT-PCR.The PCR products were cloned into the expression vector p IRES2-EGFP,which was transfected into Chinese hamster ovary(CHO-K1) cells, and then SP-ZP3 protein was obtained and purified. The highly purified SP-ZP3 protein was used in the ice-binding activity assay. The results indicated that zona pellucida 3 gene was expressed effectively in CHO-K1,and the recombinant protein was purified.Interestingly, the Zona pellucida 3 protein expressed in vitro exhibited ice-binding activity. These findings provide evidence indicating that some of fish ZP proteins may have ice-binding activity which can be evolved into cold-adaptive functions when selection pressures are present.

The reproductive cycle of clam Dosinia corrugata and related environmental factors including temperature, salinity, and Chlorophyll a level was investigated in the coast of Liaoning Province from August of 2013 to July of 2014 to understand gonadal development of the clam.It was found that the temperature and the Chlorophyll a levels were changed with seasons, and that the salinity remained the same level as the freshwater inputs were almost negligible.Histological observation revealed that the clam spawned only once a year from July to August, gametes being matured in June and July, and released collectively in July and August,as the temperature maintained in a relatively high coinciding with phytoplankton bloom or peak Chlorophyll a levels.Gametogenesis was initiated in September and appeared to continue until March, but resting phase was not observed in this study. The development of mean diameter of the clam oocyte was characterized by a clear seasonal pattern, being increased from 22.8μm in December,to the maximum value of 56.9 μm in July before spawning, and then decreased sharply. The findings indicate that the mean oocyte diameter could be taken as an effective indicator for identifying the gonadal maturity of the clam. Thus it is recommend that from May to August be the closed fishing season of Dosinia corrugata.

The morphometrical parameters including length, weight, length of antirostrum, length of rostrum, circumference and acreage of 100 sagittal otoliths were measured and analyzed in Antarctic lanternfish Electrona antarctica samples with standard length from 72.67 mm to 91.81 mm collected from the South Shetland Islands from December 2012 to February in 2013 by methods of regression analysis,Akaike information criterion(AIC) fitting model test and t test. The results showed that the relationship between the standard length and wet body weight was expressed as WW= 2.5086 × 10-5LS2.8586(R2= 0.7187,P 〈 0.05).Responding to body length, the lanternfish has large otolith, whose length was close to the width of otolith in size. The otolith has clear excisural notch and the sulcus, and the rostrum and the antirostrum are almost the same in length, although they are not obvious. The length of otolith was ranged from 3.00 mm to 3.81 mm in the left and from 2.99 mm to 3.72 mm in the right and the width of the otolith was varied from 2.98 mm to 3.62 mm in the left and from 3.05 mm to 3.67 mm in the right. The width and circumference of otolith showed stable variation with the growth of fish, however, the variation in rostrum and the antirostrum of otolith were remarkable varies for all otolith morphological parameters.A significant difference in the length of antirostrum and circumference of the otolith was found between the left and the right, but unremarkable differences in total length and acreage. The close relationship was found between the standard length of fish and the length, the width, the area, and the perimeter of the otolith. There was a linear relationship between the standard length and body weight of the lanternfish and circumference and acreage of otolith. The most stable acreage of the otolith showed a higher relationship with the standard length of the fish, with more stable growth of total length in the otolith than that of width in otolith. The findings provide the essential data with the study of age and growth of Ele

The spatial-temporal profiles and marine environments related to fishing ground distribution were investigated in albacore tuna Thunnus alalunga, one of the main catch fisheries for longline fishery in South Pacific, based on the catch data of longline fishery from 2009 to 2012 and the environmental data including sea surface temperature(SST),Chlorophyll-a concentration(Chl-a),and sea surface height anomaly(SSHA) derived from remote sensing by a generalized additive model(GAM).The results showed that the deviance of catch per unit effort(CPUE) was explained at a rate of 11.34% by the GAM,and that time(week) was the maximal contribution factor with contribution rate of 6.35%.The catching of albacore tuna was operated in the fishing grounds throughout the year, with significant seasonal change. There was higher CPUE from May to July than from January to March and from October to December. The peak fishing was observed from mid-May to later July, and the fishing grounds of albacore tuna in the South Pacific Ocean was mainly located in the water areas of 10°S-30°S and 162°E-174°E.In the central fishing ground, the suitable SST was ranged from 26.0 ℃ to 29.5 ℃,the suitable Chl-a was varied from 0.02 mg / m~3 to 0.05 mg / m3,and the appropriate SSHA was ranged from 0 cm to 20 cm.The stepwise GAM revealed that the relative importance of the six variables above in the fishing grounds of albacore tuna was followed by the order as: time(week) 〉 latitude 〉 SST 〉 longitude 〉 SSHA 〉 Chl-a concentration.
