Effects of Chinese herbal medicines Astragalus mongholicus, Glycyrrhiza uralensis and Poria cocos on non-specific immunity in Amur sturgeon

Abstract: Juvenile Amur sturgeon Acipenserschrenckii with body weight of (0.09-0.10) kg were administrated orally by Chinese herbal medicine decoction of Astragalusmongholicus, Glycyrrhizauralensis or Poriacocos at a dose of 0.30 g/kg body weight, 0.24 g/kg body weight and 0.30 g/kg body weight per day for 21 days, and the Amur sturgeon in the control group was given oral administration of distilled water. Then protein content, phagocytosis of leukocytes and lysozyme activity were determined in gill, liver, pyloric caecum, stomach, intestine and serum, sampled by caudal veins, from Amur sturgeon exposed to 30 ℃ heat stimulation for 2 h, 24 h post drug withdrawal, Amur sturgeon exposed to 22 ℃ as control, to evaluate the effects of Chinese herbal medicines on growth and blood biochemical indices of Amur sturgeon under heat stress. It was found that the maximal contents of total protein, albumin, and globulin were found in the serum of Amur sturgeon with oral administration of Poriacocos exposed to normal water temperature. There were significantly higher phagocytosis of leukocytes in the serum and relatively high lysozyme activity in the intestines in Amur sturgeon with oral administration of Astragalusmongholicus and Glycyrrhizauralensis exposed to high water temperature than those in Amur sturgeon in control group (P<0.05). The mineral lysozyme activity (0.63±0.29) μg/g was observed in the serum of Amur sturgeon in high water temperature control group, significantly lower than that in the serum of Amur sturgeon in normal water temperature(P<0.05). The findings indicated that the protein content, phagocytosis of leukocytes and relatively lysozyme activity were improved in the serum of Amur sturgeon treated by the three Chinese herbal medicines, especially against heat stress by Astragalusmongholicus.
