Abstract: Primary culture of spleen and karyotype analysis wre performed in roughskin sculpin Trachidermus fasciatus with body weight of 120-140 g by a tissue explanting method. The results showed that the optimal growth of spleen cells in roughskin sculpin was observed in the culture medium of DMEM/F12 supplemented with 20% FBS of basic fibroblast growth factor(bFGF), insulin-like growth factor-Ⅰ(IGF-Ⅰ)and chondroitin sulfate at 25 ℃ and pH 7.2. There were two types of growing spleen cells, round and oval suspended cells and cell-like and fibroblast-like adherent cells. The analysis of suspended cells revealed that roughskin sculpin had a diploid chromosome number of 40, with karyotype formula 2n=40,and 14m+10sm+16t. The findings provided a useful foundation for the establishment of in vitro cell-line and the functional investigation in roughskin sculpin spleen,and the chromosome preparation method was preliminarily established using cells of the spleen in roughskin sculpin.
