To continue researching on stress tolerance of laboratory animal green swordtail(Xiphophorus helleri) and HSP70, the cDNA partial clones encoding HSP70 protein has been first isolated in green swordtail. The isolated genes contains a consensus HSP70 motif sequence( GAT CTC GGT GGT GGC ACT TIT GAT). Comparison of the nucleotide and deduced amino acid sequences to entries in the GenBank revealed that the cDNA clone encodes a heat shock 70 kDa protein. The isolated cDNA clone nucleotide sequence exhibits a high degree of homology to the DNA sequences of HSP70 proteins from other organisms. The highest percentage of homology at the nucleotide level found to that of Japanese flounder Paralichthys olivaceus at 89%. Other fish whose homology is greater than 85% include Platichthysflesus, rainbow trout Oncorhynchus mykiss and Rhabdosargus sarba. Surprisingly, the degree of homology was even higher for the amino acid sequence of HSP70 counterpart (greater than 97% ). A significantly high degree of homology was also found with the Japanese flounder and Platichthys flesus HSP70 at 99%. The degeneracy of the genetic code allows for changes in the nucleotide sequence that do not affect the amino acid sequence of protein due to silent substitutions.

The 30 day recovery growth experiment was conducted in sea urchin (Strongylocentrotus intermedius) starved for different periods. Groups S0, S3, S6, S9, and S12 were deprived of food for 0, 3, 6, 9 and 12 days, respectively, and then refed at libitum ration levels. In the process, gonad weight, gonado -somatic index and gonad specific growth rate were observed. The results showed that there were significantly lower gonad weight and gonado - somatic index in Groups S6, S9, and S12 than those in Groups SO ,and S3. The sea urchin had maximal gonad specific growth rate at 2 or 4 days after they were refed. The histological observation showed that the structure of gonad was follicle type, consisting of germ cells and nutritive cells during growing stage in Group SO. The sea urchin starved had slow germ cell development and no more changes in nutritive cells and continued to develop after refeeding.

192 strains of bacteria isolated from the surfaces of seaweeds and 127 strains of bacteria isolated from the surfaces of invertebrates were tested for antibacterial activity by the indictors of six strains of fouling bacteria isolated from a nct cage. 60 and 49 bacterial isolates, accounting for 31.3% and 38.6% of the total tested bacteria, showed the activities against at least one target fouling bacterial strain. Different percentages of bioactive bacteria were screened from various seaweeds and invertebrates. The bacteria with bioactivity had different antibacterial spectra and strengths by the combined index. The antibacterial activities of strains P31, HF426 and AU4111 (their Index≥2.00) were found to be the highest among strains tested on the basis of the index calculated. These results indicated that the seaweeds -associated and invertebrates-associated bacteria may help the host against colonization of fouling bacteria.

The oocyte development and vitellogenesis were observed in bay scallop, Argopecten irradians concentricus under electronic microscope. Results showed that the oocyte development of the scallop was divided into three stages: previtellogenic, vitellogenic and mature oocytes; five kinds of organelles, i.e. mitochondria, Golgi vesicles, rough endoplasmic reticulum, muhivesicular bodies and mucropinocytotic vesicles were involved in formation of yolk granules, in which mitochondria and Golgi vesicles were the key organelles. Granule materials in oocytes were both endogenous and exogenous and the exogenic nutrients were mainly derived from digestive gland. It appears that the nuclear membrane takes part in formation of multivesicular bodies.

Anesthetic effects of MS - 222, clove oil and benzocaine on American shad Alosa sapidissima fingerlings were studied and two - hour transportations of the shad were conducted at the optimal anesthetic concentration. The results showed that the American shad fingerlings stopped opereular movement less than half an hour after anesthetia when the fish were exposed to concentration of MS - 222 at 75 mg/L, clove oil at 20 mg/L and bezocaine at 40 mg/L. The fish recovered to normal behaviors again after exposed in air for less than 10 rain when transferred into fresh water. The shad fingerlings had good anaesthesia for more than 12 h at 20 - 30 mg MS - 222/L, 8 - 10 mg/L clove oil and 20 - 30 mg/L benzocaine. The anesthetic effects were enhanced at high water temperature and changed with fish sizes. Small fish were more easily in good anaesthesia than the bigger one in MS -222 while the opposite case was found in clove oil and in benzocaine. There were significantly higher serum cortisols in the fingerlings 2 hs after transportation with or without anaethetics than those in the initial fish (before transportation) (P 〈 0.05). The serum cortisols in fish transported with clove oil (10 mg/L) or benzocaine (30 mg/L) were significantly lower than that in the control group (transportation without anaesthetics) (P 〈0.05 ). There was no significant differences in serum cortisol between the fingerlings 2 hs after transportation with 20 mg/L MS -222 and the fish in the control group( P 〉 0.05). The results indicated that the anaethetics especially benzocaine, had catabatic effect on the transport stress.

Nine diets containing five levels of dextrin ( 0, 10%, 20%, 30% and 40% ) and four levels of α - starch ( 10%, 20%, 30% and 40% ) , were fed to Chinese sturgeon(Acipenser sinensis) weighing (9.36 ±0.97) g for 60 days. The results showed that the amylase activity in stomach decreased while it increased in duodenum, and valvula intestine in the fish fed the experimental diets. There was no amylase activity in livers. In the diets containing different levels of dextrin, the amylase activity in stomach, duodenum, and valvula intestine reached a maximum level 5, 3 ,and 3 h after the fish fed the diets containing 20%, 30% and 10% of dextrin, respectively. The sturgeon fed the diets containing 40%, 20% and 20% of α - starch had the maximal amylase activity in stomach, duodenum, and valvula intestine 5, 5, and 3 h after feeding, respectively. There was significant difference(P 〈 0. 05 ) in amylase activity between the experimental groups. About 30% dietary dextrin is appropriate, according to the effect of different kinds and levels of carbohydrate on the amylase activity in various of alimentary canals.

Protease activities were determined in different parts of digestive organs in largemouth bass (Micropterus salraoides) at different temperature, pH and casein concentration by means of enzymological analysis. The results showed that the protease activities in different parts of digestive organs were arranged in a descent order as: stomach 〉 pyloric caeca 〉 foregut 〉 midgut 〉 hindgut. The protease activities changed with variations in temperature, pH and casein concentration in different organs. The optimum temperatures and pH were (40℃, 2.4 ) for stomach, (55℃ ,7.6) for pyloric caeca(45℃ ,8.0) for gut. The optimum temperature for protease activity was higher than that in the fish body. The highest activity of protease occurred in strong acidic condition in stomach, but in neutral or weak alkaline condition in pyloric caeca and gut. The activity of the protease also changed with concentrations of the casein, the optimum casein concentration of 20 g/L for stomach. 20 g/L for pyloric caeca, and 10 g/L for gut.

A fundamental pattern was proposed for rapid autolysis of scallop disposal from processing by a series of experiments on factors of UV radiation, mineral ions, pH value and temperature. It was found that there was the maximal amino acid nitrogen concentration (3. 389 g/L) in the scallop disposal solution under 40 min UV radiation ; good autolysis was observed at pH value of 7.0; the rate of autolysis was accelerated at 60℃ or a gradient temperature of 5℃/h from 40℃ 5 to 65 ℃ ; The rate of hydrolysis for scallop disposal increased at Na ion of 0.07 mol/L and Ca ion of 0.03 mol/L while there was no significant effect of Zn ion concentration on the rate of autolysis. The amino acid nitrogen of 4. 006 g/L was found in the hydrolysate under the optimal autolytic conditions for 3 hr. The analysis of molecular distribution of the autolyzed hydrolysates by SephadexG - 15 resin showed that the molecules primarily consisted of small molecular peptides with the molecular weight of about 784.25 Da, which can be used as essential part of the nutritious and delicate fishery seasonings.

Karuma shrimp ( Marsupenaeus japonicus ) were deprived of food for 0 ( Group 0 ), 1 ( Group 1 ), 2 ( Group 2), 3 ( Group 3 ), 4 ( Group 4), 5 ( Group 5 ), and 6 ( Group 6) days, and then refed. Approximate composition analysis of the shrimp showed the shrimp starved had low body weight and lipid levels and high ash and moisture contents 6 days following food deprivation. However, there was no significant difference in protein contents. At the end of the experiment, the body weight in each test group was similar. During the course of growth recovery, the specific growth rate was significant in variation. The food consumption increased significantly in each test group, then gradually decreased to the level of Group 0. The food conversion efficiency gradually increased on the whole. The results showed that there was completely compensatory effect in the recovery growth in the shrimp following starvation and that the compensatory effect mainly resulted from significantly increase in food consumption during the recovery growth.
