Growth,morphological characteristics and karyotype analysis of Datnioides pulcher

Abstract: Juvenile Datnioidespulcher with body mass of (0.84±0.31)g was reared in a 130 cm×45 cm×55 cm recirculating tank at water temperature of 30 ℃ and then growth performance was measured in 120, 135, 165, 182, 197, 260, and 275 days. The karyotypes were observed in metaphase chromosomes of Datnioidespulcher with body mass of 376 g and 435 g in order to understand the morphological characteristics, growth characteristics and cellular genetic background of Datnioidespulcher. The results showed that Datnioidespulcher has body flat on the side, is olive shaped, covered with small scales and complete lateral lines on body surface, with fin formula as dorsal fin (D.)Ⅺ-Ⅻ 15-17, pectoral fin (P.)16-18, pelvic fin(V.)Ⅱ 5-7, anal fin(A.)Ⅲ-Ⅳ 9-12, and caudal fin(C.)15-17. Datnioidespulcher has vertebrae of 22-24, and 4 pairs of gill arch on the each body side, and 16-19 gill rakers on the first outer gill arch. The maximal instantaneous growth rate and the best growth was observed in 165 days old fish. The correlation between body length and body mass in the juvenile stage was power index y=0.025 3x3.025 1(R2=0.991 2), which was an abnormal growth rate. The kidney cells of Datnioidespulcher were sampled to make the metaphase chromosome specimens by air-drying method. The karyotype formula of Datnioidespulcher was described as 2n=48=2m+2sm+20st+24t, with number of arms (NF) of 52, and no sex related chromosome was found, which was in the high class in the evolution of species. The findings provided reference data for germplasm standard and phylogeny of Datnioidespulcher. In present study, the morphological characteristics, growth characteristics and chromosome karyotypes of Datnioidespulcher were quantified to provide reference data for germplasm identification and phylogenetic studies.
