Isolation, and identification of pathogens and histopathological observation of cracked carapace disease in Chinese pond turtle Chinemys reevesiis

Abstract: To diagnose rotten skin and cracked carapace disease in Chinese pond turtle Chinemysreevesiis, the typical clinical symptoms were observed in 11 samples of the diseased and morbidity Chinese pond turtle collected from a breeding pond in a turtle farm in Nanchang, Jiangxi Province, and the pathogens of the cracked carapace disease in Chinese pond turtle were isolated, and identified by physical and chemical properties, physiological methods, 16S rDNA gene sequence alignment, and phylogenetic analysis. The Chinese pond turtle with cracking of the stratum corneum of the carapace and darkening of bone layer had typical clinical symptoms including sluggish and unmoving, ascites, an odor in the internal organs, dark liver, white lung, and no food in the intestine. Histopathological observation showed that the dense bone of most diseased turtles mainly lysed and necrosed, infiltrated bone marrow cavity with a large number of eosinophils, and the inflammation visceral tissue. Five bacterial strains were isolated from the carapace, liver and lung of the turtle, and the same strain was isolated from the carapace, liver and lung of the Chinese pond turtle in the artificial infection test. 16S rDNA gene sequence alignment, and phylogenetic analysis revealed that the pathogen of the Chinese pond turtle with rot skin schizont disease was identified as Aeromonashydrophila. Drug susceptibility test results showed that this bacterium was highly sensitive to cephalosporins, gentamicin, amikacin, kanamycin, and quinolone antibacterials, while it is resistant to penicillins and tetracyclines. The highly sensitive drugs above can be the first choice for clinical prevention and treatment of the disease.
