Pharmacokinetics and residues of ofloxacin in GIFT tilapia strain Oreochromis niloticus

Abstract: The metabolism and elimination of ofloxacin(OFLX)were studied in plasma and tissues of GIFT strain Nile tilapia(Oreochromis niloticus)after oral administration(10 mg/kg body weight)at(25±2)℃by high performance liquid chromatography(HPLC)method.The results showed that the distribution of OFLX in plasma and tissues were described by two open-compartment model with first order absorption with rapid absorption and distribution and slow elimination.The peak time of OFLX were found 0.41 h in plasma,3.19 h in muscle,0.18 h in liver,and 0.59 h in kidney after the oral administration.The peak concentrations of OFLX were observed to be 7.98 μg/mL in plasma,17.24 μg/g in muscle,36.10 μg/g in liver and 46.65 μg/g in kidney.There was higher OFLX level in the liver than that in other tissues and higher OFLX level in the tissues than that in the plasma at the same time.The order of the elimination was descendantly arranged as the following:kidney>muscle>liver with T1/2βof 12.90 h,19.45 h and 28.27 h,respectively.According to the maximum residue limit(MRL)of 10 μg/kg in the tissues,it is suggested that the withdrawal period be over 8 d under this experiment condition,that OFLX be orally applied at a dose of 10 mg/kg,in 12 h interval for 2-3 days when the drug is used for bacterial disease in fish.
