Construction of genetic linkage maps in largemouth bass Micropterus salmoides using AFLP markers

Abstract: The linkage maps in the broodstock and F1progeny as segregating population of largemouth bass Micropterus salmoides were primarily constructed using“two way pseude testcross”strategy with AFLP markers.A total of 2 135 markers were generated in the brood stock and 106 progenies of the mapping family by using 64 AFLP primer combinations,in which 398 markers were found polymorphic between the broodstock.Chi square analysis indicated that among the 189 female and the 209 male segregating markers,172(91.0%)female and 181(86.6%)male markers were in agreement with the Mendelian segregating ratio of 1∶1,based on the female and male genetic linkage maps constructed.The female framework map was composed of 75 segregating markers in 21 linkage groups including 7 triplets and 7 doubles,covering a total of 983.8 cM with the average interval 18.22 cM between markers. The male framework map consisted of 82 markers in 22 linkage groups including 7 triplets and 5 doublets,spanning 1 153.2 cM with the average interval of 19.22 cM.The genome length of largemouth bass was estimated o be 1 399.85 cM for the females and 1 582.72 cM for the males,and the coverage of 70.28%for the female map was observed and 72.86%for the male map.The genetic linkage maps revealed that the AFLP markers seemed to be evenly distributed within the linkage groups.
