几种鱼类的受精卵注射外源基因后孵化率的比较研究

摘要 用显微注射方法,将融合基因(鲤MT启动子基因与大麻哈鱼生长激素基因)分别导入虹鳟Oncorhynchus mykiss、鲤Cyprinus(cyprinus)carpiohaematopterus和鲫Carassius auratus gibelio 的受精卵中,注射外源基因后受精卵的孵化率分别平均为6.4%、34.0%和35.70%,按孵出的鱼苗数计算成活率,分别研究了各实验中注射后3种鱼受精卵孵化率的差异及影响因素.经t值检验,鲤与鲫注射后的孵化率无显著差异,而与虹鳟受精卵的孵化率比较有显著差异,说明显微注射在这几种鱼受精卵上的转基因孵化效果明显不同.

Abstract: By means of microinjection, fusion gene consisting of MT operator of common carp and growth hormone gene of salmon was injected in fertilized eggs released by rainbow trout, common carp and crucian carp respectively. Hatching rates averaged 6.4%, 34.0%, 35.7% respectively after injecting foreign gene. According to survival rates calculated by hatching fries, the difference and influential factors on hatching rate of these fishes were studied and analysed . There was no distinctive difference between common carp and crucian carp in hatching rate, whereas remarked differences were detected by comparing the former fishes with the rainbow trout using t test. The results indicate microinjection method can make a great difference on the transgenic effects of fertilized eggs in these fishes.
