Effects of salinity on growth rate, chlorophyll content and cell cycle of Dunaliella viridis

Abstract: The impacts of salinity on the growth rate, chlorophyll content and cell cycle of Dunaliella viridis were studied. Setting the salinity at 10, 20, 30, 40, 50, 60, 70, 80, 90, 100, the growth rate and the content of chlorophyll per liter of water were the lowest (0.121 cell/d and 908.27 μg/L) at salinity 10, whereas they were highest (0.381 cell/d and 1192.4 μg/L) at salinity 60. The changing trend of chlorophyll content per cell at different salinity was high-low-high. The minimum was at salinity 60, and the contents of chlorophyll a and total chlorophyll were 2.32×10-6 μg /cell and 3.31×10-6 μg /cell, respectively. In the cell cycle of Dunaliella viridis stage S was shortest (36.9%), then stage G2(0.8%) at salinity 60 and stage G2 shortest (0.3%), then stage S (43.6%) at salinity 80. Therefore, stage S and G2 were shortest at salinity 60-80, so was the time for cell to enter the stage of division. The results show that the optimal salinity for culturing Dunaliella viridis is 60-80.
