Skin bacterial community of wild and cultured Onychostoma barbatulum Pellegrin based on 16S rRNA high-throughput sequencing

Abstract: In order to explore the composition and diversity of the skin bacteria flora of wild and cultured Onychostomabarbatulum Pellegrin with body weight from 40 g to 70 g from different locations, the genomic DNA from the skin microorganisms of wild and cultured adult healthy O.barbatulum was extracted by CTAB method. Then the V3-V4 region of the 16S rRNA gene amplified from these genomic DNA was used for high-throughput sequencing and informatically analyzing. Results showed that the skin bacterial flora of the O.barbatulum was mainly distributed in 18 phyla, including Proteobacteria, Firmicutes, Bacteroidetes and Actinobacteria, with the top 8 genera of bacteria Hydrogenophilus, Proteus, Bacillus, Enhydrobacter, Prevotella, Epulopiscium, Cellvibrio, and Ralstonia in the richness. Functional analysis showed that these bacteria flora were mainly involved in amino acid metabolism, carbohydrate metabolism, cofactor and vitamin metabolism, lipid metabolism, secondary metabolite synthesis, and energy metabolism. Comparion of the microbial communities on skins of wild and cultured O.barbatulum revealed that Firmicutes, Bacteroidetes, Actinobacteria, Verrucomicrobia and Acidobacteria were the widespread groups of bacteria. The findings indicated that the skin bacterial composition and diversity of O.barbatulum in different environments were quite different, with the much more bacteria flora in wild O.barbatulum than that in the cultured group.
