Abstract: In order to understand the phytoplankton community structure in freshwater culture ponds,community structure of picophytoplankton (particle size 0.2-3.0 μm)and nanophytoplankton (particle size >3.0 μm)was investigated in two 4 000 m2 largemouth bass Micropterussalmoides culture ponds covered with plastic film at culture density of 3 individuals/m2 from June to September of 2018 using microscopy and high-throughput sequencing technology.The results showed that the chlorophyll a content was shown to be varied from 5.25 to 9.79 μg/L in the nanophytoplankton and from 0.62 to 2.03 μg/L in the picophytoplankton,with chlorophyll a contribution rate to the total phytoplankton of 9.8%-17.2% in picophytoplankton,significantly lower than that in nanophytoplankton(82.8%-90.2%)(P<0.05).Twenty-four genera belonging to Cyanophyta,Bacillariophyta,Chlorophyta and Euglenophyta were observed in the test ponds,with absolute dominant Cyanophyta in the four phyla,and decrease in its percent content month by month from June to September.On the premise of traditional taxonomy,only one prokaryotic picophytoplankton,Synechococcus,was found via the high-throughput sequencing,with significantly higher relative abundance in June and July than that in August and September.Except for Synechococcus,other 6 genera of prokaryotic picophytoplankton were also found in 0.2-3.0 μm phytoplankton.Eukaryotic picophytoplankton belonged to four classes of Ochrophyta,Dinoflagellata,Cryptophyta and Chlorophyta,respectively,with absolute dominant Chlorophyceae.The findings indicated that non-eutrophic aquaculture ponds were more suitable for the growth of nanophytoplankton than that of picophytoplankton.
