摘要

为了解黑边石斑鱼Epinephelus fasciatus卵巢周年发育及血清中性激素含量的变化,于2016年8月—2017年9月,在日本长崎市东中国海海域每月对其采样, 共计获得样本数量140尾,测定所获样本的体长、体质量、性腺指数(GSI)和性腺发育状态,以及血清中性激素雌二醇(E2)、性成熟激素17α、 20β双羟孕酮(DHP)和17α、 20β、21-三羟孕酮(20β-S)含量的变化。结果表明:黑边石斑鱼卵巢发育成熟的适宜水温为25~27 ℃,其GSI在2017年7—8月达到最大值,且性腺发育成熟和产卵时期也出现在此时间段;黑边石斑鱼血清中E2浓度在2017年8月显著增加(P<0.05),这表明E2对其性腺发育成熟和产卵具有重要的作用;血清中DHP浓度在2017年7—8月有增加的趋势,而20β-S浓度在2017年1月呈现峰值,此后逐渐降低,这表明在成熟时期DHP较20β-S更具有诱导性腺发育的功能。研究表明,黑边石斑鱼性腺于2017年7—8月发育成熟并产卵,可能是由于水温及性激素E2和性成熟激素DHP共同调节所致。

Abstract

Body length, body weight, gonadal development, and gonadal somatic index (GSI) were measured and calculated, and the concentration of serum sex hormones including estradiol (E2), 17α, 20β dihydroxyprogesterone (DHP), and 17α, 20β, 21-3-hydroxyprogesterone (20β-S) were detected in 140 samples of blacktip grouper Epinephelus fasciatus collected monthly in the East China Sea of Nagasaki city from August 2016 to September 2017 by ELISA (Enzyme-linked Immunosorbent Assay) aiming to understand the annual ovarian development and changes in the concentration of serum sex hormones in blacktip grouper. The results showed that the blacktip grouper had the optimal water temperature from 25 to 27 ℃ for ovarian development, with the peak GSI in July and August of 2017, and the maturation and ovulation. There was significantly high concentration of E2 in the serum of the blacktip grouper in August 2017, suggesting that E2 may play an important role in maturation and ovulation. The concentration of serum DHP was shown to be elevated in July and August of 2017. However, the concentration of serum 20β-S was found to be peak in January 2017, and then gradually decreased, indicating that compared to 20β-S, DHP was more important to induce gonadal maturation. In summary, these findings suggest that the blacktip grouper have mature gonad and ovulate in July and August of 2017, which may be attributed to the comprehensive regulation of water temperature and sex hormones such as E2 and DHP.
