Isolation of bacterial pathogen and histopathological change in cultured turbot with breaking out furunculosis

Abstract: A strain of bacterial pathogen named as DLPJC01 was isolated from the diseased turbot Scophthalmusmaximus with body weight of (180±10)g and identified as Aeromonassalmonicida subsp.achromogenes by physicochemical characteristics and PCR amplification of 16S rDNA gene. The pathogenicity of DLPJC01 to turbot was confirmed by challenge experiments. Susceptibility test results showed that this strain was highly sensitive to 5 antibiotics including gentamicin and ceftriaxone, and completely resistant to 13 antibiotics including trimethoprim, ciprofloxacin and kanamycin. Histopathological change indicated that typical acute systemic infection as observed in the diseased turbot, including the clump bacteria in all tissues checked, neutrophilic exudation in muscle tissue with necrosis of myocytes and adipocytes, acute suppurative myositis, and multifocal or diffuse necrosis with nuclear fragmentation and chromatin edging in the liver, spleen, kidney, gills and intestines as well. The findings indicated that liver, spleen, kidney, gill, intestine and muscle in the turbot were the main target organs or tissues of Aeromonassalmonicida, as a typical systemic infection.
