Effects of Buddleja lindleyana Fortune on anaesthesia and serum biochemical indices in crucian carp Carassius auratus

Abstract: Crucian carp Carassiusauratus with body weight of about 220 g was held in a 50 L tank and exposed to crude extract of plant Buddlejalindleyana Fortune at a dose of 2, 4, and 6 g/L in order to explore the anaesthetic effect of a new type of fish anaesthetic. Activities of alanine aminotransferase (ALT), aspartate aminotransferase (AST), acid phosphatase (ALP), lactate dehydrogenase (LDH-L), catalase (CAT), and superoxide dismutase (SOD) and cortisol content (COR) were determined in blood of caudal vein of crucian carp sampled by tail cutting before anesthesia (control group), just unbalanced, recovery for 0 h, and recovery for 12 h. The results showed that crucian carp was anesthetized in shortened time, recovery in longe time and had low respiratory rate with the increase in the concentration of the crude extract. The crucian carp had recovery rate of 100% within the three concentrations of the crude extract, with the most obvious anesthetic effect at crude extract concentration of 6 g/L. The blood biochemical detection revealed that there was anesthetic effect of the crude extract of Buddlejalindleyana Fortune(6 g/L) on crucian carp, with significant increase in CAT activity, indicating that the crude extract stimulated the oxidative defense reaction of hepatocytes which resisted the oxidative damage caused by substances by increasing the production rate of CAT. The findings showed that the aqueous solution had obvious anesthetic effect on crucian carp and was used as a new type of fish anesthetic.
