Effects of chromium gluconate on levels of four serum hormones and activities of glyco-metabolism-related enzymes in hepatopancreas of koi carp Cyprinus carpio

Abstract: A 8-week feeding trial was conducted to evaluate the effects of different dietary chromium gluconate levels on serum hormone levels and glyco-metabolism-related enzymes activities in hepatopancreas of koi carp Cyprinuscarpio. Juvenile koi carp with body weight of (4.18±0.38)g were reared in a plastic 300 L tanks and fed diets containing chromium gluconate at a Cr3+dose of 0(control group), 0.1, 0.2, 0.4, 0.8, 1.6, and 3.2 mg/kg with triplication. The results showed that the fish fed diets containing chromium gluconate had significantly lower serum cortisol (COR) concentration than the fish fed the control diet did(P<0.05), especially significantly higher insulin level in fish fed 0.8 mg/kg chromium gluconate diet than that in the fish fed the control diet and 0.1 mg/kg chromium gluconate diet(P<0.05), without significant difference in glycogen(GC) level in the fish fed diet containing chromium gluconate and the control diet(P>0.05). The fish fed diets containing≥0.2 mg/kg chromium gluconate had significantly higher glucokinase (GK) and lower glucose-6-phosphatase (G6P) activities than those fed the control diet did(P<0.05). The diets containing 0.1-1.6 mg/kg chromium gluconate led to significant increase in pyruvate kinase (PK) activities and to significant decrease in phosphoenolpyruvate carboxylase activities (PEPCK)(P<0.05). There were significantly higher activities of glucose 6-phosphatase dehydrogenase (G6PDH) in 0.4-0.8 mg/kg groups than that in the fish fed control diet(P<0.05). Significantly higher succinic acid dehydrogenase (SDH) activity was observed in the fish fed diet containing≥0.8 mg/kg chromium gluconate, significantly higher SDH activity than that in the control group (P<0.05), without significant difference among the other groups(P>0.05), except for Cr3+dose of 3.2 mg/kg. In conclusion, the dietary chromium gluconate shows significant impact on the carbohydrate utilization in juvenile koi carp and 0.8 mg/kg is recommended to be the optimal level under this experimental conditions.
