Optimal fermentation medium and growth in a fermentor of Pseudomonas aeruginosa inactivated vaccine in grass carp |

Abstract The nitrogen source, carbon source and phosphate were screened in culture medium of Pseudomonas aeruginosa inactivated vaccine in a single-factor test, and different ingredients of the medium were optimized in an orthogonal experiment, with a verification test of growth of the bacterium in a 5 L fermentor.It was found that the best fermentation of Pseudomonas aeruginosa in grass carp Ctenopharyngodon idellus with red skin disease was observed under the conditions of peptone+beef extract+yeast extract as nitrogen source, glucose as carbon source, K2PHO4 as phosphate source, at a dose of peptone 10 g/L, beef extract 5.0 g/L, yeast extract 2.5 g/L, glucose 5.0 g/L, K2HPO4 0.75 g/L, and NaCl 5.0 g/L.The fermentation kinetics analysis revealed that the stain had the maximal cell density at 6.44 of OD600 nm in 14 h culture, indicating that a high yield of Pseudomonas aeruginosa JP802 fermentation broth is obtained by the optimized fermentation medium.
