Comparative transcriptome analysis of body color change in red celestial goldfish Carassius auratus

Abstract: In order to explore the mechanism for body color change of the red celestial goldfish Carassiusauratus, skin transcriptome of body color development were analyzed in the 145 days old goldfish with body length of 5 cm during three different developmental stages from gray-to-orange red color by high-throughput sequencing.A total of 57.8 Gb of clean bases was obtained, and the new transcript was constructed, with 1 629 predicted new genes.Comparative transcriptome analysis revealed that 25 differentially expressed genes(DEGs)were identified in the three groups.Gene ontology functional enrichment analysis and KEGG pathway analysis indicated that DEGs were primarily distributed in the signaling pathways of cell proliferation and apoptosis, immune response,and melanogenesis.The regulation mechanism of gray-to-orange red body color transformation was preliminarily speculated in the red celestial goldfish by production practice and the comparative transcriptome analysis:in preliminary stage of the fish color change melanocyte proliferated abundantly, and then began to wither with a series of inflammatory immune response.Meanwhile, other pigment cells proliferated and differentiated until the melanocyte was finally replaced by those cells, and then the color transformation ended in the goldfish.The findings provided the references for the research on regulation mechanism of goldfish body color change.
