两种石斑鱼性腺分化前后脑垂体中FSHβ和LHβ细胞免疫组织化学及其基因表达

摘要 为了解石斑鱼Epinephelus性腺分化前后脑垂体中促卵泡激素(FSHβ)和黄体生产素(LHβ)细胞分布及基因含量的变化,以赤点石斑鱼Epinephelus akaara和黑边石斑鱼E.fasciatus为研究对象,采用苏木精-伊红染色法(H.E.)对孵化后55、80、120日龄幼鱼的性腺和脑垂体结构进行组织学观察,同时运用免疫组织化学技术(IHC)对两种石斑鱼脑垂体中FSHβ和LHβ进行识别和定位,并运用qRT-PCR技术测定其脑垂体中fshβlhβ基因含量的变化。结果表明:赤点石斑鱼和黑边石斑鱼早期性腺发育分为未分化期、分化期和卵巢期;石斑鱼性腺在55日龄时为未分化阶段,其位于肠道附近且观察到性细胞和血管,80日龄时为性腺分化阶段,性腺原基两端的结缔组织向前延伸并形成突起,表明卵巢腔正在形成,120日龄时为卵巢阶段,卵巢腔已完全形成且观察到卵原细胞,表明石斑鱼性腺已分化为雌性;两种石斑鱼在55、80、120日龄时的脑垂体结构相似,分为神经垂体(NH)和腺垂体(AH),腺垂体进一步细分为前外侧部(RPD)、中外侧部(PPD)和中间部(PI);在55、80、120日龄时,两种石斑鱼脑垂体中均检测到FSHβ和LHβ免疫信号,其主要分布在PPD区域,少量分布在RPD或PI区域;赤点石斑鱼脑垂体中fshβ基因含量在120日龄时显著高于55、80日龄(P<0.05),但lhβ基因含量无显著性差异(P>0.05),黑边石斑鱼脑垂体中fshβlhβ基因含量在120日龄时均显著高于55、80日龄(P<0.05)。研究表明,两种石斑鱼性腺在120日龄时已分化完成,脑垂体结构在55日龄时已完全形成且脑垂体中FSHβ和LHβ在两种石斑鱼性腺分化过程中起重要作用,其角色可能是促进个体的早期发育和加速性细胞的增殖。

Abstract: Follicle stimulating hormone(FSHβ)and luteinizing hormone(LHβ)cells in the pituitary were identified and localized in 50, 80, and 120 days post hatching(dph)red spotted grouper Epinephelus akaara and blacktip grouper E.fasciatus by immunohistochemistry with hematoxylin-eosin staining and the fshβ and lhβ mRNA content was detected by qRT-PCR to evaluate gonadal differentiation, pituitary structures and FSHβ and LHβ gene expression.The results showed that the early gonadal development in the two species was divided into undifferentiated stage, differentiated stage, and ovarian stage, with undifferentiated stage at location around the gut with germ cells and blood vessels at 55 dph and differentiated stage at 80 dph.The ambilateral connective tissue of gonadal primordium gradually developed forward and two slight protuberant aggregations appeared, indicating that the ovarian cavity was formed.The 120 dph red spotted grouper and blacktip grouper had ovary stage, with complete ovarian cavity and the oogonia, indicating that the sex of groupers had differentiated into female.The two species of fish had similar pituitary structures consisted of neurohypophysis and adenohypophysis which was further divided into three minor parts including the rostral pars distalis(RPD), proximal pars distalis(PPD), and the pars intermedia(PI).The immune signals of FSHβ and LHβ were mostly detected in the PPD area, with a few signals detected in the RPD or PI area at 50, 80, and 120 dph in the two species.There was significantly higher fshβ mRNA content at 120 dph than that at 50 and 80 dph(P<0.05), without significant difference in lhβ content at 50 and 80 dph(P>0.05).The blacktip grouper had significantly higher fshβ and lhβ content at 120 dph than the blacktip grouper did at 50 and 80 dph(P<0.05).The findings indicated that gonadal differentiation was finished at 120 dph in the two species of groupers.The pituitary structures were completely formed at 55 dph, and FSHβ and LHβ played important roles during gonadal differentiation.
