Abstract: In order to understand the physiological characteristics of sperm of loach Barbatulabarbatulanuda, the effects of ion concentration, glucose concentration, pH and temperature on the sperm motility of loach with body mass of (10-38 g)were investigated. The fast movement time (FT), life span (LT) and activation rate of the sperm collected from the loach induced by hormone injection exposed to NaCl at concentration of 17, 51, 68, 85, 94, and 102 mmol/L, to KCl at concentration of 5, 10, 20, 50, 80, and 100 mmol/L, to CaCl2 at concentration of 10, 30, 50, 80, and 100 mmol/L, to glucose at concentration of 10, 30, 60, 90, 150, and 180 mmol/L, to deionized water as a control, to activation solution A (the volume ratio of Na+ 68 mmol/L to K+ 80 mmol/L equal to 1∶1), or to activation solution B (the volume ratio of Na+ 68 mmol/L, K+ 80 mmol/L, Ca2+ 50 mmol/L and glucose 180 mmol/L equal to 1∶1∶1∶1). The results showed that the sperm had the strongest motility, FT of (77±23)s, LT of (495±154)s and activation rate of 95% at Na+ concentration of 68 mmol/L. The maximal sperm motility, FT of (68±10)s, LT of (412±162)s and activation rates of 95% were observed at K+concentration of 80 mmol/L. At Ca2+ of 50 mmol/L, however, the sperm motility was the highest, FT of (77±13)s, LT of (281±26)s and activation rate of 95%. The maximal sperm motility, FT of (88±11) s, LT of (272±34)s and activation rates of 90% were found at glucose concentration of 180 mmol/L. There were lower FT, LT and activation rate in Tris or glycerol solution than those in control group. The sperm did not move at all at pH of 3.0-5.0, with the maximal sperm motility at pH of 7.0, with FT of (87±11)s, LT of (128±17)s and activation rates of 90%. No significant changes in sperm motility were observed at temperature varying from 5 ℃ to 20 ℃, and sperm motility was improved in activation solution A (the volume ratio of Na+ 68 mmol/L and K+ 80 mmol/L equal to 1∶1) with FT of (91±16)s, LT of (552±90)s and sperm activation rate of 95%, and activation solution B (the volume ratio of Na+ 68 mmol/L, K+ 80 mmol/L, Ca2+ 50 mmol/L and glucose 180 mmol/L equal to 1∶1∶1∶1) with FT of (112±17)s, LT of (707±60)s and sperm activation rate of 95%. The findings indicated that 5-20 ℃ and pH of 7.0-8.0 were the suitable for sperm motility of the loach; that Na+, K+, Ca2+ and glucose enhanced the sperm motility; and the best sperm motility was observed in activation solution B.
