养殖密度对池塘循环水养殖大口黑鲈生长、生理指标和GH、IGF-I基因表达的影响

摘要

为研究池塘工程化循环水养殖模式下养殖密度对大口黑鲈Micropterus salmoides生长性能、生理指标、生长激素(GH)和胰岛素样生长因子(IGF-I)基因表达的影响,以初始体质量为(4.50±0.23)g的大口黑鲈幼鱼为研究对象,设置0.2、0.4、0.6 kg/m3 3个养殖密度组(记为SD1、SD2、SD3组),养殖试验共进行120 d,分别在养殖试验的第30、60、90、120天时,采集样本并分析各项指标。结果表明:30 d 时,SD2组鱼体质量、增重率、特定生长率均显著高于SD1、SD3组(P<0.05),而在60~120 d时鱼体质量、增重率、特定生长率随养殖密度的升高而降低;鱼体粗脂肪含量随养殖密度的升高而显著降低(P<0.05);120 d时,SD3组血清皮质醇、血糖和甘油三酯含量显著高于SD1组(P<0.05);60~120 d时,SD1组血清溶菌酶含量显著高于SD3组(P<0.05);30 d时,SD2组鲈鱼脑部GH与肝脏IGF-I基因mRNA相对表达量显著高于SD1组(P<0.05),60、90 d时,SD3组鱼脑部GH基因mRNA相对表达量显著高于SD1组(P<0.05),而SD1组鱼肝脏IGF-I基因相对表达量则显著高于SD3组(P<0.05),120 d时鱼脑部GH与肝脏IGF-I基因表达水平在各密度组间均无显著性差异(P>0.05)。研究表明,在本试验条件下,养殖前30 d,养殖密度为0.4 kg/m3组大口黑鲈生长较好,但在养殖后期,养殖密度增加会引起大口黑鲈的生长下降、免疫性能降低。

Abstract

Juvenile largemouth bass Micropterus salmoides with initial body weight of (4.50±0.23)g were reared in in-pond raceway culture systems (IPRS) which were consist of flow troughs divided into push water aeration zone, culture zone, and sewage collection area at stocking density of 0.2 kg/m3(SD1), 0.4 kg/m3 (SD2), and 0.6 kg/m3, SD3) with triplication. The growth, muscle nutrient, liver biological function and antioxidant capacity and brain growth hormone (GH) and hepatic insulin-like growth factor (IGF-1) of largemouth bass were analyzed in 30, 60, 90, and 120 days. Results showed that there were significantly higher body weight, weight gain and specific growth rate in the fish in SD2 group than those in the fish in SD1 and SD3 groups on day 30 (P<0.05). On days 60, 90 and 120, however, body weight, weight gain and specific growth rate were shown to be decreased with increasing stocking densities. The body crude lipid contents were found to be decreased with increase in stocking density, and the fish reared in the SD3 group had significant high levels of cortisol, glucose and triglyceride than those in the SD1 group on day 120 did (P<0.05). Serum lysozyme content was decreased with increasing stocking densities on days 60, 90 and 120. There were significantly higher relative expression levels of brain GH gene and hepatic IGF-I gene in SD2 group than that in SD1 group on day 30(P<0.05). There was significantly higher expressions of brain GH gene in SD3 group than that in SD1 group, while the expression levels of hepatic IGF-I gene were significantly lower in SD3 group than that in SD1 group on days 60 and 90(P<0.05), without significant difference in the expressions of GH and IGF-I genes on day 120 (P>0.05). In conclusion, largemouth bass reared at stocking density of 0.4 kg/m3 had better growth and health status before days 30, and after that, the fish reared at high stocking density in IPRS inhibited the growth and reduced immune function.
