Phagocytosis and agglutination of coelomocytes in sea cucumber Apostichopus japonicus with relation to water temperature

Abstract: Phagocytosis and agglutination of coelomocytes in sea cucumber Apostichopus japonicus with the relation with water temperature were studied by light and electron microscopes. The phagocytosic assay showed that Spherolocytes and Amoebocytes had the abilities to phagocytize garmine and yeast, and the abilities of phagocytosis were correlated to the kind and size of the foreign bodies. In the assay of phagocytizing garmize, the maximal phag- ocytosis (48.01%) was observed 6 hours later, and 108 hours later the phagocytosis was over. In the assay of phagocytizing yeast, the maximal phagocytosis (35.17%) was occurred at 4 hours later, and the phagocytosis was over 96 hours later. Furthermore, the type III Spherolocyted only phagocytize garmine. In addition, the function of phagocytosis was correlated to temperature. The peak phagocytic percent and index appeared at 18℃, which was higher at 12℃ than that at 4℃ and 25℃. The coelomocytes had the abilities to agglutinate in vitro and the agglutination was correlated to the stimulation by temperature and foreign bodies.
