裙带菜孢子叶褐藻聚糖硫酸酯分离树脂的筛选及抗氧化活性研究

摘要 为探究不同树脂对于裙带菜Undaria pinnatifida Suringar袍子叶褐藻聚糖硫酸酯粗品的分离纯化效果及分离纯化后各组分的组成和抗氧化能力,以酶解法制备的裙带菜孢子叶褐藻聚糖硫酸酯(UPS-FUC)为原料,选取11种不同种类的树脂,通过静态吸附和动态洗脱试验筛选对其分离效果最佳的树脂,以维生素C(VC)为对照物,研究了5个组分的多糖对于羟自由基(·OH)和超氧阴离子自由基的清除能力,采用苯酚-硫酸法和明胶-氯化钡法分别测定纯化各组分的总糖及硫酸根含量,采用高效液相色谱法测定各个纯化组分的单糖组成并测定各分离组分的抗氧化活性。结果表明:通过静态吸附和动态洗脱试验,确定了分离效果最佳的树脂为D315;当其上样液的pH为8.0时,分离纯化后可得到5个组分的多糖,其回收率分别14.90%、11.45%、17.21%、13.54%、12.67%,各组分多糖的含量分别为69.95%、63.47%、46.98%、47.80%、44.45%;各组分对两种自由基均具有一定的清除能力,其抑制·OH和的最低IC50值分别为F1组分的1.13 mg/mL和2.95 mg/mL,且其抗氧化活性与组分中多糖的含量呈正相关关系。研究表明,弱碱性阴离子交换树脂D315在上样液pH为8.0条件下分离效果最好,其分离得到的5个组分中F1的抗氧化效果最好,且各组分的抗氧化活性与岩藻糖和半乳糖的含量呈正相关关系,本研究结果可对裙带菜袍子叶褐藻聚糖硫酸脂粗多糖的分离纯化及其活性研究提供参考依据。

Abstract: In this study, the optimal separation resin of crude fucoidan (UPS-FUC) prepared from sporophyll of macroalga Undaria pinnatifida Suringar by enzymatic hydrolysis was screened from 11 kinds of resins by static adsorption and dynamic elution experiments, the total sugar content and sulfate content of each component was determined by phenol-sulfuric acid method and gelatin-BaCl2 method in order to explore the separation and purification effect of different resins on the crude fucoidan sulfate of the macroalga. The monosaccharide composition of each purified component was determined by high performance liquid chromatography, Fenton reaction detection method and o-benzene. The antioxidative activity of each purified component was determined by triolol oxidation to evaluate the composition and antioxidant capacity of each component after separation and purification. The results showed that the best separation resin was D315 and five components were separated and purified by D315 resin from the fucoidan at the optimum pH of 8.0, with the yield of 14.90%, 11.45%, 17.21%, 13.54% and 12.67%, respectively,and the mass fractions of polysaccharides of 69.95%, 63.47%, 46.98%, 47.80%, and 44.45%, respectively. The analysis of scavenging ability of the five component polysaccharides for hydroxyl radical (·OH) and superoxide anion radical  revealed that each component had a certain ability to scavenge both free radicals when vitamin C(VC) was used as a control. The minimum IC50 value of fraction 1(F1) was 1.13 mg/mL for inhibition of ·OH and 2.95 mg/mL for inhibition of  with proportional antioxidant activity to the content of polysaccharides in the components and the fucose and galactose contents. The findings provide a theoretical reference for the separation, purification and activity study of crude polysaccharides from the sporophyll of the macroalga Undaria pinnatifida Suringar.
