Abstract

The intestinal microflora structure was comparatively investigated in healthy hybrid snakehead Channa maculata(♀)×C.argus(♂)with body mass of (1 140.67±45.88)g cultured in soil ponds (abce123, abce124, and abce125) and healthy hybrid snakehead of body mass of (1 130.00±96.27)g cultured in high level membrane ponds(abce126, abce127, and abce128) by the high throughput sequencing method in order to probe into the differences in the intestinal microflora structure of the hybrid snakehead in two culture modes. The results showed that there were significantly lower contents of total ammonia nitrogen, nitrite nitrogen, nitrate nitrogen and total alkalinity in the high level membrane ponds than those in the soil ponds, without significant difference in water temperature and pH values under the two culture modes. Obvious differences in the microflora structure in the intestine were observed in the hybrid snakehead in different culture modes, with much higher diversity of bacteria in the intestinal tract of the hybrid snakehead in the soil ponds than that in the high level membrane ponds. At the level of phylum, the Proteus in abce123 had significantly higher percentage than other category bacteria did, with the maximal percentage of Proteobacteria in abce124 and Bacteroidetes in abce125. There was the maximal percentage of Fusobacteria in the three intestinal samples in high level membrane ponds. At the genus level, however, all unknown dominant bacteria were found in the intestinal samples in the soil pond abce123. ChryseobacteriumMycoplasmaAcidovoraxAcetobacter and Cetobacterium were of the dominant bacteria in intestinal samples in the abce124 according to the abundance. The dominant bacteria in the intestinal samples were ChrysobacteriumAcinetobacterAcidophilusMycoplasma and Cetobacterium in abce125 based on the abundance. The most abundance of Cetobacterium, accounting for more than 95%, was observed in the three samples in the high level membrane ponds. The composition and diversity of the intestinal microflora were quite different in the hybrid snakehead under the different cultivation modes. The findings provided theoretical basis for the selection of the cultivation mode and the microecology research of the hybrid snakehead in the future.
