Re-immersion physiologic recovery of yesso scallop following different stress treatments in the quality determined period

Abstract
The experiment was performed in two post-harvest stages including a quality determination period(QDP)corresponding to on-board handling and post quality determination period (P-QDP)corresponding to land-based depuration treatment in order to explore the relationship between the post-harvest stress and the storage stability of the live scallops. First, the live yesso scallop Patinopectenyessoensis with shell length of 110 mm landing on the lab was immediately held for 24 h as the scallop with initial physiologic state (I). Secondly, the held yesso scallop was divided into two groups for 36 h QDP stress treatment, emersed (E) and emersed with mechanical shocking (free falling from 1 meter height to the concrete floor) (E-S), and sampled at 0 h, 3 h, 6 h, 12 h, 24 h, and 36 h. Finally, the yesso scallop in E and E-S groups were re-immersed into circulating sea water system for 3 d P-QDP recovery, and immersed treatment was carried out as control(C). Live scallops were evaluated using sensory and physico-chemical indicators including pH, and contents of glycogen, phosphoarginine (ArP), and ATP-related compounds, and adenylate energy charge (AEC) value. It was found that the live scallop showed an efficient stress relief after 24 h holding, with significant increase in contents of ATP and glycogen and AEC value. The analysis of physic-biochemical characteristics indicated that damages including mantle shrinkage were observed in the yesso scallop to some extent in both E and E-S groups, with the worst in the yesso scallop with length of 9 mm. There is no significant difference in the physic-biochemical indicators between E group and E-S group at the end of the QDP(P>0.05). However, the yesso scallop in E-S showed more insensitive reactivity responding to needle stimulation comparing to the scallop in E group. After re-immersed for 3 days in the P-QDP, the scallop in E and E-S groups had significant difference in the recovery(P<0.05). The yesso scallop in E group recovered from QDP had life indicators near to the initial physiologic state with a very sensitive response to external stimuli; while the scallop in E-S groups was unrecoverable and maintained at the same insensitiveness in the QDP. In summary, there is a quality determination period during post-harvest, which impacts a delayed influence on quality and shelf-life of live scallop.
