Abstract: In this study,RNA extract in liver of taimen Hucho taimen was used as a template,and IGF-Ⅱ gene open reading frame was amplified by one step RT-PCR using primers designed by Atlantic salmon IGF-Ⅱ gene indexed by Gen Bank(NCBI No.NM_ 001123647) to evaluate role of fish insulin-like growth factor-Ⅱ(IGF-Ⅱ) in the development of taimen embryos.Bioinformatics analysis revealed that the IGF-Ⅱ contained an ORF of 645 bp encoding 214 amino acid with a signal peptides and five regions named as B,C,A,D,and E.The IGF-Ⅱ of taimen had molecular weight of 24 554 and the isoelectric point of 9.92,shared high identity(98.6%) with salmon and the homology of the IGF-Ⅱ gene at nucleotide and amino acid level.The IGF-ⅡmRNA expression was detected in different tissues using Real-Time quantitative PCR technique.It was found that the maximal expression was observed in the liver,followed by in the heart,gill,spleen,hindgut,kidney,stomach,and muscle,and the minimal expression in the foregut and brain.
