一株黄金鲫鱼源蜡状芽孢杆菌的筛选、鉴定及耐性研究

摘要 为筛选出适合制作水产微生态制剂的黄金鲫Carassius auratus鱼源益生菌,从黄金鲫肠道分离细菌,通过产酶能力测试、体外抑菌试验和抗生素敏感性试验进行筛选,筛选菌株经纯化培养后通过形态特征观察、生理生化及分子特征进行鉴定;采用紫外分光光度计和稀释涂布平板计数相结合的方法,建立细菌活菌数与OD值的线性曲线,测定细菌对高温、酸性、胃液、肠液、胆盐耐受能力;通过固定黄金鲫体外肠道黏液蛋白模型,结合荧光标记物Hoechst 3325标记细菌法,检测细菌对黄金鲫肠黏液蛋白的黏附能力,并通过急性毒性试验检测菌株的安全性。结果表明:从黄金鲫肠道中共分离出5株具有产酶能力的细菌,其中HJJ-1菌株具备较好的产酶特性,可有效抑制金黄色葡萄球菌和大肠杆菌的生长,且对多种药物敏感;进一步对该菌株进行鉴定及益生效果评价,16S rRNA基因序列分析显示,HJJ-1株与蜡状芽孢杆菌Bacillus cereus的一致性达100%,结合形态观察和生理生化试验结果,最终鉴定HJJ-1菌株为蜡状芽孢杆菌;菌株OD600 nm值(x)与活菌数(y)的回归方程式为y=5.493 3e7.823 8x(R2=0.990 2),该菌株具有良好的耐高温、耐酸性、耐胃液、耐肠液、耐胆盐等特点,可黏附于黄金鲫肠道黏液蛋白,且安全性良好。研究表明,本试验中最终筛选出的黄金鲫鱼源蜡状芽孢杆菌性能优良且安全,具有潜在的益生特性。

Abstract: Bacteria were isolated from the intestine of Golden crucian carp Carassius auratus and identified by morphological observation, physiological and biochemical analysis, and molecular characterization. The linear relationship between the number of viable bacteria and OD value was established by UV spectrophotometer and counting method, and the tolerance of bacteria was evaluated by measuring the tolerance of bacteria to high temperature, acid, gastric juice, intestinal fluid and bile salt. The adhesion ability of bacteria to intestinal mucus protein of golden crucian carp was detected by fixing the intestinal mucus protein model in vitro and combining with the fluorescent marker Hoechst 3325. The safety of the strain was tested by an acute toxicity test,in vitro antibacterial test, and antibiotic sensitivity test in order to screen out golden crucian carp-derived probiotics suitable for the production of aquatic microecological preparations. It was found that 5 strains with enzyme-producing ability were isolated from the intestinal tract of the golden crucian carp, in which HJJ-1 strain had good enzyme-producing characteristics, effectively inhibited the growth of Staphylococcus aureus and Escherichia coli, with sensitive to a variety of drugs, so the strain was further identified and the probiotic effect was evaluated. Sequence analysis of 16S rRNA showed that HJJ-1 was 100% similar to Bacillus cereus, combined with morphological characteristics and physiological and biochemical indices and finally identified HJJ-1 as Bacillus cereus. The regression equation between OD600 nm value and viable count of the strain was expressed as y=5.493 3e7.823 8x (R2=0.990 2), characterized by characteristics of high temperature resistance, acid resistance, gastric juice resistance, intestinal fluid resistance, bile salt resistance, adhesion to the Golden crucian carp intestinal mucus protein, and good safety. The findings indicated that Bacillus cereus isolated from golden crucian carp intestines had good potential probiotic characteristics and an excellent and safety.
